Abstract

MnFe2O4 nanoparticles are used in medicine for cancer treatments through hyperthermia, gene, and drug delivery systems. The primary purpose of the current investigation was to study the effect of the MnFe2O4 nanoparticles on the structure and function of acid phosphatase. The enzyme activity displayed that MnFe2O4 nanoparticles acted as a non-competitive inhibitor. Ki was calculated to be 0.217 mM. The thermal denaturation analysis also demonstrated that Tm was decreased by adding MnFe2O4 nanoparticles concentrations (from 342.0�338.5 K). Far-UV circular studies also revealed that MnFe2O4 nanoparticles could alter the secondary structure of acid phosphatase by the enhancement of the α-helix contents (from 10.8 to 11.3) and the decrease of the β-sheet (from 30.2 to 29.8). It was also shown that MnFe2O4 nanoparticles decreased the intrinsic fluorescence of acid phosphatase via the static quenching. Also, with the rise of temperature, the Stern�Volmer constant was declined from 9.95 � 10 4 to 2.42 M� 10 4�1. The hydrogen bonding and van der Waals interactions also played a significant role in this interaction. These results also showed that MnFe2O4 nanoparticles changed the conformational of acid phosphatase. Overall, MnFe2O4 nanoparticles could influence the activity and stability of acid phosphatase.
